چکیده

Yeasts utilize tryptophan for the synthesis of nicotinic acid under aerobic, but not under anaerobic, conditions. Under aerobic conditions of growth, radioactivity from tryptophanJ4C uniformly labeled in the benzene ring and 14C!-carboxyl-labeled 3-hydroxyanthranilic acid is incorporated into nicotinic acid. Under anaerobic conditions, little radioactivity from these compounds is incorporated into nicotinic acid. The presence of a tryptophan-nicotinic acid pathway in yeast is further supported by the establishment of the presence of formylkynurenine formamidase and d-hydroxyanthranilic acid oxidase, two of the enzymes known to mediate the conversion of tryptophan to nicotinic acid in mammalian systems. Under anaerobic conditions, yeasts incorporate radioactivity from uniformly labeled aspartateJ4C and glutamateJ4C, suggesting that an alternative pathway similar to that shown in several bacterial systems is operative. Evidence is presented which establishes quinolinic acid as a precursor to nicotinic acid adenine dinucleotide under either aerobic or anaerobic conditions. Quinolinic acid appears, therefore, to be a common intermediate in the alternative pathways for nicotinic acid biosynthesis in yeast.

Precursors of nicotinic acid in Escherichia coli.

It has now been firmly established that mammals and Neurospora are able to convert tryptophan to nicotinic acid via a biosynthetic pathway which contains several well characterized intermediate compounds (2). However, evidence has been accumulating which indicates that in bacteria the biosynthesis of nicotinic acid is accomplished by a different pathway. Volcani and Snell (3) demonstrated that ...

Saccharopine, an Intermediate of the Aminoadipic Acid Pathway of Lysine Biosynthesis. Ii. Studies in Saccharomyces Cereviseae.

Previous studies with wild yeast strains (Torulopsis utilis and Saccharomyces cerevisiae) have provided circumstantial evidence for the involvement of oc-aminoadipic-6-semialdehyde (1, 2) and saccharopine (3) in lysine biosynthesis from or-aminoadipic acid. Evidence drawn from appropriat’e nutrit.ional studies, accumulation of aminoadipic acid metabolites, and enzymatic analysis with a series o...

L-Asparagine auxotrophs of Saccharomyces cerevisiae: genetic and phenotypic characterization.

L-Asparagine auxotrophy in Saccharomyces cerevisiae is the result of mutation in each of two unlinked cistrons, ASN1 and ASN2. Mutation in only one of these cistrons yields growth indistinguishable from that of wild-type cells under a variety of nutritional stresses. Relatively high concentrations of L-asparagine are required to permit maximal growth of the auxotrophs, and the amino acid requir...

Biosynthesis of Zinc Oxide Nanoparticles using Intracellular Extract of Saccharomyces cerevisiae and Evaluation of its Antibacterial and Antioxidant Activities

Introduction: Attention to the biosynthesis of nanoparticles (NPs) has been increased recently since they are cost-effective, eco-friendly, and potential alternatives to chemical and physical methods. This study aimed to synthesize zinc oxide nanoparticles (ZnO NPs) using an intracellular extract of Saccharomyces cerevisiae. Moreover, it was attempted to evaluate their antibacterial and antioxi...
